Summary:
Animal bodies exhibit intricate cellular organization, forming tissues, organs, and systems under precise morphogenetic guidance. While achieving molecular-level reconstruction of complete cell types and morphogen gradients throughout the entire animal is essential for comprehending pattern formation during development and regeneration, it remains unrealized. We construct a comprehensive molecular profile of animal bodies by employing spatial transcriptomic reconstruction across regeneration timepoints in planarians, unveiling the dynamics of regeneration-responsive spatial domains and genes at the molecular level. We decode the spatial gene expression patterns underlying positional gradients along the body axes, tracking their changes throughout regeneration. Our study captures complete cell types and positional information in animals, elucidating the regulatory principles of scalable, self-organized regenerative patterns potentially universal across diverse phyla.
Overall design:
PRISTA4D database has a total of 370 coronal sections from CIW4 planarian uninjured animal and regenerative animals at 0 hpa, 12 hpa, 36 hpa, 3 dpa, 5 dpa, 7 dpa, 10 dpa and 14 dpa, with each stage represented by two animals. For each individual, serial sections were collectted and profiled with Stereo-seq transcriptome sequencing. Digital 3D whole-body animals were re-constructed.
