Summary:
The mechanisms underlying the interaction between disseminated tumor cells (DTCs) and their tissue microenvironment during metastasis colonization are poorly understood. We integrated multimodal single-cell and spatial profiling from liver cancer mouse models and human metastases to track the spatiotemporal dynamics of DTCs and their microenvironments across nine sequential time points from single-cell seeding until overt lung metastasis.
Overall design:
We established a stable model of lung metastasis in immunocompetent mice by intravenously injecting mCherry-labeled Hepa 1-6 murine hepatocellular carcinoma (HCC) cells (derived from a female mouse) into male C57BL6/J mice. Whole lungs were collected at nine post-injection time points spanning from initial cancer cell seeding to metastatic outgrowth: T1 (0.25 h), T2 (1 h), T3 (4 h), T4 (12 h), T5 (24 h), T6 (2 days), T7 (8 days), T8 (21 days), and T9 (35 days). At each time point, maximal right lung cross-sections were cryosectioned at 10 µm thickness to capture a roughly single-cell layer of tissue for spatial enhanced resolution omics-sequencing (Stereo-seq).
