Summary:
In this study, by employing the spatial enhanced resolution omics sequencing (Stereo-Seq), we have successfully generated a comprehensive atlas of single-cell spatial transcriptomes in potato leaves that were inoculated with P. infestans at four different time points during the early infection stages. Our findings collectively contribute to a comprehensive understanding of the spatial and cellular responsive atlas in plant-pathogen interactions, thereby offering valuable targets for the effective management of potato late blight.
Overall design:
We collected leaf samples from diploid potato A6-26 plants that were inoculated with P. infestans strain 88069td (expressing a cytosolic tandem RFP protein) at 3 hours post-infection (hpi), 6 hpi, 12 hpi and 24 hpi. The potato leaves were inoculated with water (mock) at each time point and served as the control group. Cryo-sectioned potato leaves from each time point were placed on four separate chips. To capture the transcripts at a single-cell level, we conducted fluorescent brightener (FB) staining of the plant cell walls and oligonucleotide staining of nucleus on the chip to visualize cell-cell boundary. Subsequently, performed single-cell segmentations. The transcripts were assigned to individual cellular regions based on the coordinate information of the cell wall boundary to obtain a spatial transcriptome atlas with single-cell resolution.
