Summary:
The intrauterine environment, composed of the placenta, fetal membranes, and the maternal uterus, undergoes intricate and significant changes during embryonic development. However, it has not been systematically characterized in a spatial view, which limit our understanding of placental development and maternal-fetal diseases. Here, we used Stereo-seq to establish the comprehensive transcriptome landscapes on 13 segments of the mouse uterus at 6 different time points ranging from E7.5 to E14.5, integrated with previously published scRNA-seq data. We identified the specific regional composition of the ectoplacental cone and elucidated the spatial trajectory of trophoblast differentiation during early post-implantation stages. Our atlas of the intrauterine environment revealed interface-specific TFs at the early interface of placental invasion and uncovered spatially restricted communications in the labyrinth. Furthermore, we revealed the distinctive spatial distribution of maternal and paternal imprinting genes, as well as genes associated with heart development, providing valuable spatial datasets especially for congenital heart disease. Importantly, our study demonstrated that maternal high-fat diet, as an adverse intrauterine exposure, upregulates inflammatory responses and disrupts angiogenesis regulation in the interface of placental invasion. Overall, this study has established the most systematic spatiotemporal single-cell resolution transcriptome atlas of the intrauterine environment placental development following embryo implantation.Overall design:
MPSTA database has a total of 15 sagittal sections from C57BL/6 mouse uterine at E7.5 (x2), E8.5 (x2), E9.5 (x2), E10.5 (x3), E10.5 HFD(x1), E12.5 (x2), E14.5 (x2), E14.5 HFD (x1) using Stereo-seq. For each stages, at least two sections were included.Platform:
