Summary:Hair loss and grey, the earlier visible signs of skin aging, are due to the impairment of the hair follicle stem cells and their niches. Combined with single-cell RNA sequencing and spatial transcriptomic technologies, we studied the age-related transcriptional patterns associated with age in 57,181 cells from 11 scalp samples. We identified three subtypes of mitotic keratinocytes which localized in the inter follicular epidermis (IFE), outer root sheath (ORS) and hair matrix by deconvolution in the spatial chip and confirmed that they were in the transition stage by pseudo-time trajectory analysis. In aged scalp, AP1 transcription factor complex (such as JUN, FOS, FOSB, JUNB) were activated, DCT gene was upregulated in melanocytes by cell-type-specific analysis, BMP and ncWNT signaling was decreased in the intercellular communication between dermal papilla (DP) and hair follicle keratinocytes. These finding provided a cell-type-specific transcriptional clues of the aging process of hair follicles and may assist in future hair follicle regeneration therapies.
