In this study, we present a Gut Phage Biobank (GPB), a collection of 104 isolated gut phages obtained through a refined workflow. We investigate their genomic and biological characteristics, explore the phage-host interaction mechanisms, pose potential application for bacteria inhibition both in vitro and in vivo, and develop a promoter toolkit based on gut phage-bacteria pairs. The GPB and the synthetic biology toolkit offer a valuable resource and a novel, comprehensive perspective on gut phages.
