Project name: DNA sequencing of Medicago truncatula from ChIP-seq experiments of Embryos at different developmental stages using the H3 pan Acethyl and the H3K27me3 antibodies
Description: We analysed the presence of H3 pan-acethylation and H3K27me3 histone marks bound to DNA of Medicago (A17) at different stages of seed development from seeds produced at 20°C and 26°C.Overall design: Regarding the H3 pan-acetyl marks, we performed ChIP-seq experiments on two replicates of at least 50 embryos of each stage/replicate (S1, S2) from seed produced at 20 and 26 degrees. Regarding the H3K27me3marks, we performed ChIP-seq experiments on one replicates of at least 50 embryos of each stage/replicate (S1, S2, S3 and S4) from seed produced at 20 and 26 degrees. The four selected stages correspond at before (S1) and after (S2) desiccation tolerance acquisition and before (S3) and after (S4) longevity acquisition in dissected seed embryo tissues of seeds produced at 20 and 26 degrees. Corresponding stages S1_20°C=17DAP, S2_20°C=26DAP, S3_20°C=36DAP, S4_20°C=44DAP, S1_26°C=14DAP, S2_26°C=17DAP, S3_26°C=22DAP, S4_26°C=28DAP (DAP=days after pollination)
