Description: We used a genetic screen in Saccharomyces cerevisiae to generate novel spontaneous mutations that confer resistance to 2-deoxy-D-glucose (2DG), a toxic glucose analogue with anti-cancer potential that is known to disrupt glycolysis. One resistance-conferring mutation in hexokinase-2 arose in five independently evolved strains. Curiously, though this mutation does encode a loss-of-function allele, the affected amino acid does not appear to interact directly with bound glucose/2DG.
