Description: We report targeted RNA-seq of Ribosomal Protein Subunit 12 mRNA transcripts in two editing complexes, RESC and REH2C, and in total mitochondrial RNA. Accurate editing is enriched in RESC-associated RPS12 and requires the helicase complex REH2C acting in trans. The initiating gRNA directs very accurate editing in the first few positions. However, two-terminal adenines in the initiating gRNA direct a non-canonical edit that causes pausing in editing progression. REH2C also controls this pausing in editing.
