Project name: Sinorhizobium meliloti 2011

Description: Sinorhizobium meliloti 2011 WT bacteroids of Medicago sativa proximal nodule sections methylation detection; Sterilized seeds were germinated on Kalys agar in the dark. Seedlings were inoculated with the bacteria, and then transferred to pots containing a perlite:sand mixture. Plants and grown for 39 or 40 days in a greenhouse with occasional watering with a 1 g L-1 nutrient solution (PlantProd solution [N-P-K, 0-15-40; Fertil]). At the end of the growth period, nodules were harvested, manually sectioned at the pink-white interface, and stored at -80 until further use. Bacteroids were enriched from the nodule material using Percol gradient centrifugation prior to DNA isolation.
