Description: In this work, we characterized the mRNA transcriptome, small RNA transcriptome, and degradome profiles of 35S::PS in relation to the wild type tomato. In particular, we comprehensively annotated tomato miRNAs and identified miRNAs differentially regulated by systemin. These efforts were supplemented with detailed genetic studies of two specific miRNAs - miR157 that is induced by systemin and miR828 that is suppressed by systemin. We found that the differentially expressed miRNAs are both sufficient and necessary for the pleiotropic developmental phenotypes observed in the 35S::PS plants. These results provide holistic insights in the regulatory networks in tomato integrating peptide signaling and miRNA action.
