Project name: Expression analysis of S. pimpinellifolium accession LA1589 wild type, lc, fas and lc/fas NILs, SlCLV3 RNAi and SlCLV3-complementation lines during tomato floral development.
Description: The goal of this study is to decipher the effect of the lc and fas mutations on gene expression during floral development in tomato as well as determining the effect of strongly downregulating tomato CLV3 using the RNAi strategy. The strand-specific RNA libraries were sequenced through Illumina HiSeq2000 at Genomic Resources Core Facility at Weill Cornell Medical College for 50 bp single–end sequencing. Total raw reads for each sample ranged between 20-30 millions.Overall design: Tissues were collected from mature plants grown in the greenhouse of wild type, lc, fas, lc/fas NILs, RNAi-SlCLV3 lines and SlCLV3 complementation lines in the fas mutant background at three developmental stages, each with four replicates. The first stage included inflorescence meristems, floral meristems and 1 to 3 days post floral initiation (dpi) floral buds. The second and third stages included floral buds at 4 and 6 dpi, respectively.
