Description: GmMEKK1 silencing was performed in Glycine max (Williams 82) using virus-induced gene silencing (VIGS) mediated by Bean pod mottle virus (BPMV) by inserting a 360-bp fragment of GmMEKK1 in the BPMV2 vector. RNAseq analysis was performed to compare the transcriptional profile of GmMEKK1-silenced plants vs vector control plants. The fully expanded second trifoliates from top were collected at 2 weeks post inoculation when the silencing phenotype was evident. The RNAseq analysis had 4 biological replicates. Each replicates contained leaf samples from 3 independent plants of both the GmMEKK1-silenced and vector control plants, respectively. Total RNA was extracted from each replicate sample using Qiagen RNA extraction columns and genomic DNA was removed subsequently by DNaseI digestion. The resulting RNA samples were used for constructing 8 multiplex libraries and sequencing was performed at the National Center for Genome Resources on an Illumina Genome Analyzer II.
