Description: To isolate resistance genes from tobacco, a high quality tobacco cultivar (Nicotiana tabacum cv CB-1) was treated with a varieties of bio- and abio-stresses, such as different hormones and various of adversity situations; then, cDNA libraries were prepared from extracted RNA of various treated samples and the controls and subjected to 454 XLR70 Titanium Pyrosequencing, abtaining 309903 sequences reads from treatments and 290953 reads from the control. After assembling and removing redundancy sequences by blasting Uniprot and the 26 main plant species EST unigene databases including that of tobacco in the GenBank, 38810 unigenes were obtained with the ContingN50 reached 782bp. The 38810 unigene were then subjected to removing of redundancy sequences by blasting the available and the most complete tobacco unigene database with 114917 sequences on SGN, we acquired 16231 new tobacco unigene finally.
