Transcriptome sequencing of diverse bio-geographical accessions of barley

Description: Total RNAs were extracted using RNEasy plant mini kit (QIAGEN GmbH, Hilden, Germany) from leaves and roots of 6 barley (H. vulgare) accessions and a wild barley (H. spontanium),according to the manufacture's instruction. The barley plants used for RNA extraction were grown by hydro- culture in a growth chamber. The growth chamber were maintained at 16-h light: 8- h dark at 22?C. Poly(A)+ RNAs were purified using MicroPoly(A)Purist Kit (Life Technologies, USA) according to the manufacture's instruction. Libraries for stranded-RNA sequencing using an Ion torrent sequencer were constructed using Ion Total RNA-Seq Kit v2 (Life Technologies, USA) according to the manufacture's instruction with the Ion Xpress Barcode Adapters (Life Technologies, USA) corresponding to each accession. Templates for 200 base-read libraries sequenced by the Ion Proton semiconductor sequencer (Life Technologies, USA) were prepared using Ion P1 Template OT2 200 Kit v3 (Life Technologies, USA), Ion P1 Sequencing 200 Kit v3 (Life Technologies, USA) and Ion P1 Chip Kit v3 (Life Technologies, USA), according to the manufacturer's instructions. The sequencing run was carried out using an Ion Proton sequencer (Life Technologies, USA) with an Ion P1 chip.
