Description: A Dehalococcoides containing culture dechlorinating cis-1, 2 dichloroethene to ethene via vinyl chloride was enriched from the sediment mud of lotus field. Microcosms were set up on the medium containing cis-1,2 dichloroethene as electron acceptor and hydrogen and lactate as electron donor. Finally, the culture that degrades 10ppm of cis-1, 2 dichloroethene via vinyl chloride in 14 days was obtained. PCR amplification and sequence analyses of Dehalococcoides 16S rRNA gene and reductive dehalogenase gene exhibited that the culture contains a Dehalococcoides strain with vcrA, vinyl chloride reductive dehalogenase gene of Dehalococcoides sp. VS. The metagenome was applied to the genome analysis by next generation DNA sequencers, SOLiD 3 and PacBio RS. The full length genome of Dehalococcoides sp. was reconstructed from the contains of PacBio RS data. The genome sequence was confirmed using the data of SOLiD 3. The strain was named as Dehalococcoides mccartyi IBARAKI.
