Description: The goal of this study is to compare differences in alternative splicing between stimulated and restimulated ex vivo isolated CD8 T cells.Overall design: Naïve (CD44- CD62L+) CD8 T cells from the spleen of 5-6 weeks WT mice (n=3) were sorted using a BD FACSAria with 5 lasers (BD Bioscience). 105 naive CD8 T cells were stimulated in vitro with 4 ug/ml of purified anti-mouse CD28 and 20 ug/ml of plate-coated, purified anti-mouse CD3 in an incubator at 37ºC and 5% CO2. Recombinant human IL-2 (10 U/ml) was added to the culture at the end of day 1. After 3 days of culture, cells were left untreated or washed and additionally restimulated for 1 hour using a Cell Stimulation Cocktail, PMA/Ionomycin. In vitro activated CD8 T cells were then washed and resuspended in 1 ml TRIzol.
