Description: Overall design: BCL11B knock-out hESC line was generated using the CRISPR/Cas9-assisted genome editing technology with guide RNAs targeting exon 2 of the BCL11B gene. To investigate whether BCL11B deficiency affects the homeostasis and function of human striatal and cortical neurons, we performed striatal and cortical neural differentiation on the BCL11B-/- line along with the isogenic control HUES9 iCas9 cells using an established protocol (Arber et al., Development 2015). Total mRNA from control and BCL11B-/- neurons was collected and prepared for RNA-seq analysis at day 30 in vitro neural differentiation.
