Project name: ChIRP-seq of lincDUSP in colon cancer cells
Description: The genome-wide occupancy of lincDUSP was determined by using chromatin isolation after RNA purification to detemine the genomic sites where lincDUSP directly interacts with chromatin.Overall design: Colon cancer cells were grown to obtain over 500 million cells. After crosslinking with formaldehyde and sonication, half of the lysates were incubated with probes that bind to lincDUSP, and the second lysate was incubated with control non-specific probes over night. Purification of lincDUSP, DNA isolation and sequencing enabeled us to identify the genome-wide occupancy sites of lincDUSP.
