Description: To investigate the effect of RBPMS KO on mRNA ribosomes occupancy, polysome profiling was performed on lysates from WT and RBPMS KO hESCs.Overall design: Ribosomes were stalled on mRNAs by using cycloheximide and ribosomal fractions were isolated using polysome profiling. RNA was isolated using Trizol and equal amounts were used for library generation for stranded, paired-end RNA sequencing.
