Description: Background:Poly(I:C) is a synthetic double-stranded RNA that is often used as a substitute for dsRNA viral infection, and previous studies mainly focused the role of endometrial epithelium in immune response and mucosal barrier against dsRNA virus. Stromal cell is critically important for function of endometrial layer. However, whether endometrial stromal cells (ESCs) directly produce immune-response to the virus, and the biomolecules, long non-coding RNA in ESCs involve in immune-response still is unknown. In this study, we detected the immuno-response status in vitro cultured ESCs after Poly(I:C)-induction, and then analyzed mRNA and lncRNA expression profiles to explore the role of these RNAs in the immune response.Results:291 lncRNAs and 1311 mRNAs were significantly differentially expressed (DE) between the control and Poly(I:C) groups (p<0.05). GO and KEGG analysis showed that DE genes and target genes of DE lncRNAs were enriched in many pathways related to immune and inflammatory responses, such as the toll-like receptor and the NF-κB and Jak-STAT signaling pathways. Co-expression analysis of lncRNAs and mRNAs revealed seven novel lncRNAs and their 78 targeted genes enriched in immune response pathways. In particular, the two novel lncRNA MSTRG.39626.1 and LncRNA MSTRG.137189.4-targeted genes widely involved in immune-related signaling pathways. The results suggested that the novel lncRNA MSTRG.39626.1 and MSTRG.137189.4 in the ESCs might be involved in extensive regulation of the immune response to Poly(I:C) through genes related to immune signaling pathways.Conclusions:Our results provide both transcriptomic and epigenetic insights into the immune response of ESCs to dsRNA virus infection.Overall design: Poly(I:C) stimulated or not stimulated rabbit endometrial stromal cells were analyzed by RNA-seq, in triplicate.
