Description: uncultured Termite group 1 bacterium phylotype Rs-D17.The wood-feeding termite Reticulitermes speratus (Isoptera; Rhinotermitidae) was collected in Saitama, Japan. The entire gut was removed from a worker termite, and a single cell of the cellulolytic protist Trichonympha agilis (Parabasalia; Trichonymphida) in the gut was physically isolated by using a micromanipulator. In order to collect the intracellular symbionts of Trichonympha agilis, as-yet-uncultivated bacteria designated phylotype Rs-D17 belonging to the candidate division Termite Group 1, the host Trichonympha agilis cell was ruptured in buffer and bacterial cells which leaked out were collected with a glass capillary. These collected cells were directly subjected to isothermal whole genome amplification. After checking their purity by clone analysis of 16S rRNA gene and its adjacent ITS, the complete genome sequence was determined using ABI 3730 capillary sequencers and a GS20 system.
