Project name: In vitro mixture of eight SARS-CoV-2 samples
Description: A mixture of eight single-patient samples that originated from oropharyngeal swabs collectedin January 2021 in Slovakia (GISAID identifiers and Pango lineages EPI ISL 959634 B.1.1.7, EPI ISL 959635 B.1.1.7, EPI ISL 959642 B.1.160, EPI ISL 959631 B.1.258, EPI ISL 959649 B.1.258, EPI ISL 959636 B.1.258, EPI ISL 959648 B.1.258, EPI ISL 959647 B.1.1.170). Sample processing and sequencing library construction were carried out at the Biomedical Research Centre of the Slovak Academy of Sciences in Bratislava, Slovakia. Individual samples were diluted according to the quantification cycle (Cq) to obtain approximately equimolar mixture. This mixture was then used for reverse transcription, followed by PCR amplification using the 2-kb primer scheme by Resende et al., 2020 and sequenced using ligation kit (SQK-LSK109) on R9.4.1 flow cell (FLO-MIN106) and a MinION Mk-1b device (Oxford Nanopore Technologies). Nanopore sequencing data were base called using Guppy v.4.4.1.
