Transcriptome sequencing of Bactrocera dorsalis female adults after mimic-318 treatment

Description: To identify potential target genes regulated by miR-318 in Bactrocera dorsalis, transcriptome libraries were constructed from female adults injected with 50 pmol mimic-318 on day 3 post-eclosion. Total RNA was extracted 24 h post-injection using the method described in the RNA and DNA isolation section. RNA concentration and purity were determined using a NanoDrop 2000 spectrophotometer (Thermo Scientific), and RNA integrity was evaluated with the RNA Nano6000 Assay Kit on an Agilent 2100 Bioanalyzer (Agilent Technologies, Santa Clara, CA, USA). Sequencing libraries were prepared from 1 ug total RNA per sample using the Hieff NGS Ultima Dual-mode mRNA Library Prep Kit for Illumina (Yeasen Biotechnology, Shanghai, China), and sequenced on an Illumina NovaSeq platform to generate 150-bp paired-end reads, with a sequencing depth of 6 Gb per sample.
