Description: Based on Sc2.0 synthetic yeast and Synthetic Chromosome Rearrangement and Modification by LoxP-mediated Evolution (SCRaMbLE), low selenoprotein synthesis efficiency and selenium toxicity and other key problems in selenoprotein yeast synthesis will be solved. we will reveal the impact of beneficial targets on the metabolic network and redox regulation, and finally establish the yeast genome modification principle for efficient synthesis of selenoprotein. Meantime, we will explore the molecular mechanism responsible for the loss of selenoprotein synthesis machinery in yeast during evolution.
