Description: We have generated naive cells from ESCs and MYCN/BCL2 overexpressing ESCs cultured in 4CL culture system (named 4CL and 4CL/N/B). Single-cell RNA sequencing was used to analyze the 4CL gene expression pattern and the effect of MYCN/BCL2 overexpression on cell pluripotency and cell cycle. We observed 4CL were positioned homogeneously closer to embryonic day5 (E5) of the human pre-implantation blastocyst. 4CL/N/B ESCs had a transcriptional profile enriched in naive pluripotency genes similar to 4CL ESCs. Besides, enrichment of cell cycle related gene ontology terms for upregulated genes in 4CL/N/B and a higher percentage of cells entering the G2/M phase in 4CL/N/B ESCs compared to 4CL alone in this analysis.
